Here's the situation:

I use my internal hard drive to store my music. I have an external hard drive that I split with Time Machine & Movies. I am running out of room on my external hard drive and want to buy a new one. I still want to use the old partition to store my movies as well.

Can I have an iTunes library in two separate locations? How would I do that?
 
When the following highlighted field is unchecked, you can have your music on as many drives as you want, as the iTunes Library only stores the location of your files.

Your imported and copied music is stored under iTunes Music, but your iTunes Library file just stores the location.
