My iTunes library is currently residing on a NAS that runs 24h and is directly connected to the iMac I am using.

In iTunes' preferences, I have set the folder on the NAS as the 'official' media folder. However, this setting is reset to a standard folder in the user directory (/Users/username/Music/iTunes/iTunes Media), even though there is no music stored there.

Why does iTunes do this (maybe due to the NAS not being immediately available after resuming from sleep?) and more important, how can I fix this?

I think your theory about the resuming from sleep might be the culprit. I have had my iTunes library on my NAS since Leopard days with my Mac Mini and now my iMac and never had the problem you describe, but I don't have the Macs or the NAS ever go to sleep.

To test your theory, turn off the sleep feature for a day and see what happens.
