iTunes not letting me transfer videos in my phone. help required

hello everybody
i just bought iphone4. I'm new user. my problem is when i try to transfer video file from my PC to phone. iTunes don't even show that file in que to sync it to phone.I'm using Handbrake most latest version to encode HD video files for iphone4.and another thing is it also not allow me to "check for updates" and " Restore " as both options are disabled..... what could be the reason ?
my baseband is 1.59.00 and OS is iOS 5.0.1

i also tried DISKAID to transfer my video file to phone, i copied my video file to camera folder ( where all camera pics and video recordings are saved ). but still nothing....

is quicktime compulsory for itunes in order to transfer Handbrake converted video files to my iphone ?
