I have un-ticked (removed) iTunesHelper from Login Items. However each time I restart Mac, it appears again in the Login Items. I don't want iTunes Helper to open automatically when I log in.

What can I do ?

Unchecking it does not remove it, it just hides or unhides it. Select it and hit the minus button to remove.

That's exactly what I did. I removed it by selecting the (-) button. But after a restart, it appears again, indicating that Mac opened it automatically.

I'll test on mine after work. I'd search the forum for iTunes helper , I remember this during the Lion DP. I think it is launched when iTunes is open but not sure why is added to log in items.

The iTunesHelper is now gone. What I noticed was, when iTunes is launched - iTunesHelper pops up in the Login Items.

Is iTunesHelper important? I disabled it but I don't want to mess up my new Mac...
 
There should be an option in iTunes preferences to open iTunes when device connected (I'm not on osx right now so I don't know where it is exactly)
If you turn off that, I think iTuneshelper will not reappear in login items.

I'm afraid I'm not seeing that option available on latest iTunes :(
 
Perhaps iTunes Preferences > Devices > tick "Prevent iPods, iPhones and iPads from syncing automatically" will do what you want...

It's already checked, but helper is still running...

The iTuneshelper.app is part of the iTunes package. To diable the iTuneshelper.app you have to view the contents of the iTunes.app by Option clicking, or right clicking, and choosing "Show Package Contents". The iTuneshelper.app is located in the MacOS folder.

Rename iTuneshelper.app to iTuneshelper-disable.app and this will prevent it from launching and being added to Login Items.

This method tested and working with OS 10.8.3 and iTunes 11.0.2.
