Hello, I'm planning on unlocking my current iPhone 3GS 32 GB and selling it when I buy my iPhone 4 on launch day. If my current phone isn't jailbroken or unlocked or anything right now (runs iOS 4) then will it lock up when I activate my new iPhone 4, keeping it from being able to jailbreak? In other words, do I need to do the unlocking and everything before I buy my iPhone 4?

Activating your new iPhone won't do anything to your current iPhone. Activating the new microsim will cause the old sim to get kicked out of the system, but that won't do anything to the phone.
