According to Kevin Rose - a mockup of iTunes 8 is shown here. So the on the left hand side you get genius playlists, which might replace or accompany the old smart playlist area. Genius sidebar on the right hand side (sounds similar to the mini-store recommendations).
Another change apparently being that the 3 views offered for music are
List, Grid, Cover flow versus list, list with artwork, coverflow. If it is true that the grid view will show album covers in an array and offer mouseover playback controls, it would seem to be a move towards the mouse over play capability being touted in Snow Leopard.

A slider like iPhoto/ pictures in mobile me to control the album artwork size in grid view also. Whether Genius will be looking at style, bpm, or other to match it's unsure. Presumably it can recommend from your own collection as well as other artists and songs you don't currently have. (Pandora, musicplasma etc spring to mind).

Other changes include the addition of this visualisation, and alongside iTunes, some iPod action.
