If I install Lion using an USB-stick is it possible to do an upgrade or is only a clean install possible?

You cannot go from Leopard to Lion as Lion requires some a few updates from 10.6.8 to install, and how would you even get it as Leopard doesn't have the App Store?

I've got a Snow Leopard Macbook Pro, so I'll 'burn' the image to an USB-stick and use that to upgrade the Leopard iMac :)

Lion doesn't require 10.6.8 to install, in fact it doesn't require any OS at all to install. And you can always purchase Lion on some friend's Mac that has App Store on it, copy it onto a thumb drive and install it from there.
 
You can only upgrade if your running 10.6.8, otherwise the installer wont run. You will need to perform a clean install

That's a shame :), but, can I use my Leopard Time Machine disk to restore my files and settings then?
