Can the Home Button be deactivated or locked from accidental pushing? I have a wallet type case and when I carry it in my back pocket it's turning the screen on, and sometimes activating voice dialing.

No. BTW your title is misleading... seems like you're about to shoot your homebutton and are telling it to watch out - which would make no sense.
