On my external drive I installed 10.5.8 on it and I've only used it with my macbook pro for booting up. Is it possible to plug it into, for example, a mac pro and run the partition or does it only work with one machine?

If you have an operating system on an external drive you should be able ta use it to boot multibable machines.
 
Yup, it can be used to boot multiple machines. Just be wary on booting up a version of Mac OS X that is earlier than what a particular Mac model first ever shipped with as it may lack drivers, etc.
