Maybe Iam confused, the 500gb 16mb are a double speed read of the 8mb of 320gb? or its the same speed read?

Rotation speed is the same (7200rpm). Reading speeds can be different as one HD has more platters than the other (not by much). Buffer size (as you asked) won't make a difference for speed. As rule of thumb remember: a bigger drive will be faster for a longer time than a smaller one.
