Hello!
I am now a proud owner of a Macbook Pro 15 2012 non retina 2.6! :D
Got it today and I wanted to do a small bit of Minecraft on it so I plugged it in on my first boot so I could use the Nvidia 650 in it. The Mac says its fully charged. Does this mean I can unplug it now?
Also, can I use the Nvidia 650 on battery and get about an hours life out of it?

You can run with it plugged in or unplugged. It will not overcharge. Run on battery whenever you need to and plug it in whenever you can. You can plug or unplug at any time, regardless of the charged percentage. Just make sure you don't run on AC power all the time, as your battery needs to be used regularly to stay healthy. This should answer most, if not all, of your battery/charging questions:
