Macbook memory upgrade/backup question

Hello

This is my first post so please bare with me, when it come to technologic speak I have no idea..

I have a few questions.

I have a late 2009 MacBook with 2gb of memory. But I'm wanting to upgrade to 4gb so I can upgrade to Lion.

Firstly, do I need to back-up everything before I upgrade my memory? Can I do this by using Time Machine? If so, will it all fit on my 8gb memory stick?
