I will be in a dark room and the light iwll only come on once I adjust them. I have the setting checked in my system preferences.
 
Is it possible that your display's brightness is high enough that the light from the display tricks the keyboard's sensors into thinking you've got enough ambient light?

I've seen something similar mentioned in the past.

But when the OP walks into a dark room, the ambient light sensor should automatically change the display brightness.
