mobile me sync vs usb sync

I'm confused when to sync via Mobile me and with iTunes via usb.

I have mobile me running pretty well. I'm getting info pushed around seemingly fine. I do have podcasts I want to get, etc. If I sync my iPhone via iTunes/usb, will that change anything with my contacts/calendar/email sync between Mobile me, my Mac, and my iPhone???

In iTunes under iPhone / Info - Make sure to TURN OFF Sync for the things you do not want to Sync. Like Contacts, Calendar etc...
