I am wondering how many will be using this to track their kids or spouses?

If someone has 3 phones on a family plan can all 3 be registered with the program or will each phone need its own account? Just curious.

I bet that it is associated to one MobileMe account. There may be a MobileMe preference pane in iPhone OS 3.0 though in which case you could tie multiple phones to the same account. I'm interested in seeing how this is going to work as well.
 
Could you sign up for the trial of MobileMe if you lose your phone, to find it? Assuming you have the IMEI number? Or do you need to have the iPhone in hand?

No, you have to setup your MobileMe account on your iPhone and then enable the option in the settings to allow the Find My iPhone feature to work properly.
