My wife is upgrading from her 3G to my iPhone 4, can her number be transferred from home or do I need to go to AT&T to do that?

You need a new microsim, so that means a trip to an AT&T store. There's always a chance you could call customer service and ask them to mail you one, but it'd take days and their first suggestion would be to just go to a corporate store anyway.

So the microsim that's in the 4 can't be "redone" with her info? My 4S came with a new microsim which I transferred to over the phone.

Not as far as I know, AT&T has always said the old sim once deactivated is dead. Can't be redone to a new number.
