After updating to 10.14.6, my "I" key has become unresponsive. Sometimes it will work, but most often times it requires me to press it multiple times for it to finally register on screen.

Anyone else?
 
Which machine? If its a modern apple notebook it is probably just the (at this point, standard failure of the) keyboard, and just a coincidence that it's started to happen the same time as your macOS update.

For what it is worth, my I key works fine on my 2015 running the most recent macOS.

Late 2012 rMBP.

Never had any issues whatsoever with the keyboard until the latest update.

You will hardly find anybody who will not agree with throAU that updates do not cause malfunctions in keyboards.
It is no doubt a timely coincidence.
Software changes can disrupt the work of a computer but in an entirely different way.
 
Try a PRAM reset. Turn your machine off, hold command + option + P + R and while holding, turn on the machine. It'll chime, restart, then on the next chime (after restart) you can release the keys.
