I am thinking about getting one of these 2FA authentication keys for extra security. These mostly have the NFC function is when you just need to tap on the back of a mobile device to get authenticated.

Question is, does the iPhone 12 Mini support NFC? I looked through the setting page and could not locate any NFC controls.

What if I don’t choose the NFC option? Will Apple Mail app ask me every time to insert the key just to check my emails?

Because there are versions of yubikeys that have a lightning connector on the other end along with a USB-C connector at the other end of the key.

99% of the time I use Apple Mail app on the iPhone to check emails.

If I need to insert the key each time to check emails, then it would be very annoying.

It's app-specific or site-specific whether or not a security key can be used. To the best of my knowledge, Apple Mail does not support the use of security keys.

For those places that do support security keys, most (perhaps all) give you the option to stop requiring the use of the key after the first time you authenticate with the key. In other words, you can mark a device as "trusted."

Here's Yubico's list of sites and apps that support the use of Yubikeys. It's not necessarily up to date.

If you do decide to use a Yubi Key with a password manager or browser etc., you really need to buy two keys. If you only have 1 key and it get lost or damaged, you could end up locked out of your account. In my opinion, it is best to set up a software token (from the hardware key setup) if the app you are using supports such. That way, you are less likely to get locked out.
 
What about the Outlook app on my iPhone? Is that supported?

If so, then I will use the Outlook app. But having to insert the key into the phone each time would be a hassle.

Outlook does support 2FA - and Microsoft, Google and RSA both have RSA software token apps - the latter uses it’s own keys connected to server implemented mostly by corporations, so RSA would not help.

The other two will work. You need to check your service provider regarding what 2FA apps are supported.
