Does a hp all-in-one need any special activation at the time of purchase in order for it to operate normally once you get it home and set it up?
 
No, it shouldn't be required. I have seen POS activation for Microsoft Office 365 offered in places like Office Depot, but it usually has a free trial period out-of-the-box during which the user can activate it at home. I have not seen any such POS activation for Windows 10.

My advice: Demand an explanation if a retailer insists that activation is needed. If the answer seems iffy, shop elsewhere.
