I have a 42 mm, SS Apple Watch 2 and am getting an Edition, gray Apple Watch 3 next week. I know that when pairing the watch to my iPhone, I'm going to be given the choice to "Restore from Backup" or "Set up As a New Apple Watch." What is the difference between these options, and which one should I use, considering that I want this watch to utilize cellular service?

If you want the new watch to have the exact same apps and setting without you have to do that all ever again, then do a restore from backup. Also if you want to switch between two watches.
My personal advice is always setup as a new Watch, less incompatible issue so usually the software will run more smoothly.
