If I make a partition via disk utility (from startup disk), and I decide I don't want to keep it later on, will I have to reformat the drive in order to remove the partition, and revert it back to one partition?

unless you use iPartition, yes you will have to reformat. It may be possible to do it using terminal commands, but I have been unable to ascertain exactly how.
