Picking between 512GB or 1TB SSD. Does Dropbox work on portable hard drive?

I'm buying a new 15 rMBP for work use. I need to run 300GB of files on Dropbox at all times. Just wondering if Dropbox works on portable hard drive? Or if Dropbox would only sync properly if installed on the main SSD?

Shouldn't be a problem as long as it is formatted in HFS+

Dropbox, as in the actual application, must be on the start up drive but you can make the Dropbox folder (the data) point to the external hard drive.
