Q about video quality, visualhub and mkv converting

Should I use the video flag "Best possible H.264/AVC" in conjunction with the apple TV 5.1 + 2.0 , "go nuts," and 2 pass encoding to get the best looking video?

I don't really care about file size at all. I'm just wondering if the "Best Possible H.264/AVC" flag helps the video look better.
