I have an external drive with all of my music and home movies on it. iTunes reads all of that content from the drive. If I rename the drive will it effect playback? For instance, if I change the name of the drive from "A" to "B" and then try to play a song in iTunes, will iTunes say it can't find the song?

I'm pretty sure the consequences won't be pretty.
Much like changing the name of your home folder will screw your system up pretty badly.

You can conduct a little experiment though, by using a USB stick or something like it, and reenact the scenario you explained.
 
After renaming the drive you can start iTunes whilst holding down the option key, iTunes will then prompt you to select a new location for your iTunes library.
