I have a USB harddrive and am wondering whether I can repartition it from MS-DOS(FAT32) to a format that will hold my video files from iMovie.

Sure, just use Disk Utility

Yes. Open up disk utility with it plugged in, select the drive, and you can make a new partition with the Mac OS Extended (journaled) format, then delete the old partition. And just the regular OS Extended should work just as fine.

Or an easier way to do it would be to just erase the drive with a different volume format.
