Should I go for SATA-2 or SATA-3? Does the DVD connector support SATA-3?

Depending on your model, it supports either or only S-ATA 1.5 Gbps (S-ATA I), if you have a 2007 MBP for example.
Check System Profiler > Hardware > Serial-ATA and see for yourself.
Some 2011 MBPs have S-ATA 6.0 Gbps (S-ATA III) interfaces for the ODD.

Is there a reason why you wouldn't want to put the SSD in the existing hard drive bay (that may be SATA-III)? You can then transfer your hard drive to the optical bay with a mounting bracket. My hard drive didn't connect at 6.0Gbps anyway when it was in the hard drive bay.
