No. You don't need to be plugged into power to access Siri on the 6s. It has an always-on M9 chip.
I'm just wondering if this will only work when the screen is on or if it will extend to when the screen is off as well.
 
This is actually a good question... will Siri work when the phone is locked? I'm guessing you don't want someone who stole your phone be able to access Siri and your info as well?
 
In one of the videos they activate Siri whilst it was across the room, most likely locked.

Well at the moment you can already access Siri when your phone is locked? Unless you disable the ability to do so. But I watched a preview video on YouTube of the iOS 9 GM yesterday, and when enabling the 'hey siri' functionality it asks you to say a few statements in order to recognise your voice. However if you just used the home button you could probably still access Siri no matter what your voice is!
