I don't know if this has always been an issue, or if it just happened in iOS 14.5....

On iOS for iPhone and iPad (but not MacOS), if Siri's language is set to English (United Kingdom) with the voice as either Irish Voice 1 or Irish Voice 2, the letter "a" seems to be missing.... ask Siri to spell, e.g. banana, with the above voice settings, and it is spelled "B N N". Is this just me or does anyone else have this issue?

I have Siri’s voice set to British Voice 1, and the letter ‘a’ is announced when spelling words. I switched to Irish Voice 2 to test for you and, indeed, it misses the letter ‘a’ when spelling out words. (This was tested on iPadOS 14.5.)
