I have over 40GB of my 250GB Macbook Pro storage space taken up by 'Other'.
Does anyone know what this could be?
It's too big to just be the recovery partition, and I've checked the largest files on my MBP are movies, as you might expect.

Any ideas?

The reporting in System Profiler is wonky. Noticed the same thing. I would check with DaisyDisk or diskring.

Used DaisyDisk, a lot of it was made up from Library, Hidden, Private & System.
When you say System Profiler being wonky...what's the solution?
