I already ordered MBP 15" early 2011.

The main bay is apple 128SSD. I am planning to buy one more hd for the storage. But what is the difference between with a second hard drive in the Optibay and external drive ???

I mean any difference with the speed ? As I know, Optibay will use sata2. And the external drive will be select FireWire 800...

Or any difference I need to concern ?

The Sata will be slightly faster... Even though FW800 talks about having a speed of 800mb/s its not entirely accurate. The difference is not big, but enough for to choose the optibay solution. I think you'll notice the biggest difference when dealing with larger files.
 
My external FW800 drive does around 65MB/s reads and writes. Your SATA-II harddrive can do, if it's a fast one, maybe 90-100MB/s.

USB 2.0 does around 35-38MB/s approx.
