My iMac's (OS 10.8) 1TB internal hard drive is now filled to the brim with mainly iPhoto libraries. I would like to move the photo libraries to a USB external drive. Then for backup, I want to backup the internal and this USB drive with the photos on it to a 2nd USB external drive. Is this possible using Time Machine?

Yep... external drives are by default excluded from Time Machine backup. So after you attach the drive with the photos go into Time Machine prefs and under Options tab you will see the external on the exclude list. Just minus it out of there and it will be included in the backups.
