Have some new headphones I'm trying to break-in by running it overnight on iTunes. Have done this many times up to a couple years ago. The problem exists whether I have Automatic Login turned on or off.

2011 13" Macbook Pro
10.10.4

Computer setup:
- to never sleep
- allow display to sleep
- "Put Hard Disks to Sleep", unchecked
- check all my music (much more than 24 hr playback)
- start iTunes

What happens is when I go back to it a few hours +/- later:
- keyboard backlight is on
- the display is on sleep
- system is logged out
- iTunes has quit

Which version iTunes?
Are you running Apple Music, iTunes Match, Genius or other such stuff? Are you connected to internet overnight?
What happens when you tell iTunes to repeat the same song all night long?
