I am about to purchase an iPhone 4S off of craigslist for my wife to upgrade her from the 3GS. What do I need to do about the regular SIM card to the micro SIM? Do I got to AT&T to get a micro SIM? How do I get her number over to the new phone?

Propably best to do it at an AT&T store after you get the phone.
 
Apple Stores in the U.S. have AT&T micro-SIMs. If you explain what you're doing they might give you an unactivated micro-SIM for free. Once you get the 4S, call AT&T and explain that you're upgrading and you already have the micro-SIM. They'll make note of the ICCID and deactivate your wife's regular SIM card and activate the micro-SIM. I've done this multiple times.
 
Meet the person at an AT&T store so they can be sure everything is OK before you buy it. You don't want to end up buying a stolen phone or one that doesn't work properly.
