I have the Apple Mail program set up to use with my AOL Mail account. I notice that AOL Mail has an option for two-factor authentication. So if I were to log into AOL Mail directly it would prompt for a security code sent as a text message. I'm wondering whether I could make use of this option while still using Apple Mail.

Is Apple Mail logging into AOL Mail each time I click on Get Mail, or is it always logged in based on the password I entered when setting up the connection to AOL Mail? Is anyone using Apple Mail with AOL Mail and making use of two-factor authentication?

Not speaking from experience with AOL, but how it may work is: turn on two factor, then generate a password for Mail. Mail will reject your existing password, then ask you to enter your password. Here, you'll enter the newly generated one, which should keep Mail authenticated for future use.
