And if so, how significant? I have an original 500GB Time Capsule, but it doesn't seem to cut it anymore, with up to 6 wi fi devices on it at once.

Would a new Airport Express be an upgrade over a first-gen Time Capsule?

Definitely a significant upgrade. Simultaneous dual-band support, antenna improvements, range increased by a good 40% as compared with a 1st gen TC.
Why not pick up the latest AirPort Time Capsule instead? AirPort Express + network storage. You can't go wrong.
