Can I set up an external hardrive to my Airport Extreme and have a wired connection to it as well? This way I could have a firewire connection to it on my iMac and a wireless connection to it for my Macbook?

That would be a nifty way to go. I suspect the answer is no, but I do not know for sure. I'm thinking the drive controller in the external case would probably freak out if it were getting multiple requests at the same time.
 
A standard home use external hard drive can't be used by both connections at the same time - either AESB or wired.

Same reason you can't plug the same hard drive into 2 different computers at the same time.
