The '1' will be when Tim Cook is presenting, Steve calls him from home, Tim reaches in his pocket and pulls out the brand new redesigned iPhone 5, and Steve introduces it through FaceTime.
It means iPhone 5 will be so amazing that every time you look at it you will stand in shock from amazement and miss all your calls.
The map most certainly does mean something. The stuff on the Maps icon is not just random roads and a number, it is the actual (simplified) layout of Apple's campus. 280 is Interstate 280, which runs through Cupertino right by Apple HQ. Hence the Maps icon on the invitation tells us where the event is.

Right. But it's important to note that that's the standard Maps icon and has been for a while. Check the iPhone.
