I've been curious: When an update comes out, would there be any difference updating my phone OTA vs via iTunes sync? I know this has been mentioned, but I didn't quite understand.

No. If update has some number, then same. I do OTA.

iTunes will download the full installer while OTA will only download update data. For example, the full install may be a 2GB file while OTA update will only be a few hundred MB in size.
