For example, USPS, UPS, etc give me an estimated time of arrival in business days. Does the day the package is shipped count as a business day? Same for the day it is being delivered.

Well a business day is Monday - Friday.
So lets say UPS says it will be delevered in 3 business days. The package shipped Monday, so it should arrive Thursday (Tuesday, Wednesday, & Thursday).

It also depends on what time the package is dropped off at the shipping facility of choice. IIRC it is before 3 p.m. at most places.
So how long would it take for a USPS First Class envelope to reach NJ from PA when shipped before 3PM?

They don't guarantee any times for First Class. If you use a ZIP+4 it supposedly will get there on the 2nd day, but it still isn't guaranteed.
