Yes you can. It'll work straight out of the box, you won't have to set any options.

I know, because I'm using the controller myself to play Far Cry 2 and it works flawlessly.

GTA IV will very likely support a 360 controller from the get-go. If not there will certainly be a mod for it, as there was for GTA SA to support a 360 controller.
