I have an Airport Express 802.11N with latest software (7.4.1). It has stopped connecting for AirTunes.
In the Airport Utility (from a PowerBook G4) the music icon has a cancel ("O" with "\") superimposed.
The Airport Utility from an Intel-based MacPro or MacBook Pro shows the icon properly. In all cases the Enable AirTunes block is checked.
iTunes shows the Airport Express in the list but will not play through it. It has worked before.
 
if i have problems connecting to my express, I've found that resetting my router will help. (the main one that serves the G network that the express connects to). typically i just leave the express alone

every so often mine hangs on "connecting to..." and the reboot tends to fix it.
 
Have you tried connecting headphones to the output jack on the Express to see if you get any audio whatsoever?

As far as the PowerBook having the funky no symbol there, did you update its AirPort Utility to 5.4.1?
