I'm almost 100% sure its tomorrow, June 21st.

iOS 4 on the 21st, iPhone 4 on the 24th.
Does this mean since it's the 21st on the east coast Apple will release the new iOS or will they wait until it's the 21st over there, 3am on the east coast? I remember for one of the updates they released it during the middle of the night. Just wondering if anyone thinks they will do that?
Well, to tell you the truth, I don't think a few hours makes that big of a difference. Although about your question: I'm not sure, sorry.
they will release during the day most likely i say from 9am PDT- 2pm PDT. just my guess
iOS 2 and iOS 3 were both released mid morning/early afternoon...
Don't expect the update to come possibly until 12 hours from now
If I had to guess, I would say that the update will be available in the late morning/early afternoon on the East coast.
That's just my guess based on what myself and others have experienced in the past.

According to this article, last year it was around 9 AM PDT.
However, with iPhone 2.0, it was 5 AM PDT (Source) . (Although it was a really messed up day for anybody wanting to access Apple's servers.)
