Once I put a file in iTunes (like from my "Users>mrat93>Movies" folder) and now it's in both in "Users>mrat93>Movies" and "Users>mrat93>Music>iTunes>iTunes Music>Movies" folders, do I need to keep the file from "Users>mrat93>Movies"? Thanks.

Do you mean can you play your files in any other applications? Otherwise your question makes little sense. 😱
 
I meant maybe a shortcut to the actual movie in my "Movies" folder so it doesn't take up space.
 
No, and I assume that you have backed-up the original file to some other disc anyway. Back up your data before you lose it!
