My wife currently has one Dell P2720DC. I would like to upgrade her to dual monitors - either the Dell U2723QE or Dell P2723QE (I can't really tell the difference between the two).

Anyway, I know her current monitor is 2K and either of the new ones would be 4K. One monitor will be the main monitor but will it be annoying for her nonetheless to go back and forth between the 2 resolutions?

In my experience no, not at all. I use a 5k iMac next to a 2k Eizo. Eizo only for photography. Both screens are 27" and next to each other . Maybe depending on personal usage (?).

I have different model Dell monitors but they are physically the same size, look and bezels. They are also both 1440s.

It would drive me nuts if they looked different side by side but not sure how I would feel about same look but different resolution, probably a non-issue. YMMV

I've had slightly different size and resolution monitors before. I absolutely hated it. Some people don't care and others do. If she's kind of OCD about things being neat, tidy, squared away, balanced, &c. It'll probably be annoying for her.

The most annoying thing will be if there is a noticeable scale difference. So, even if both are set to the same scaling. Pictures, text and cursor visibly jump in size. Also if the mouse cursor jumps up or down when moving from one screen to the other.

I bought three of the same make and model monitors, with VESA mounts. I use a triple monitor stand to make everything nice and neat. Plus reclaim desk space.

Same monitors a must? Not really. Choose best for purpose. For many, it's one bigger main display and one smaller secondary for a special purpose (video previews, photo editing, coding, Teams/Slack/Discord, etc). For me, I mostly use 'office' type apps and often need to work on multiple documents/windows at a time, so I chose two identical monitors so things look the same from one display to the other. Again, best for purpose, and that meant two of the same for me.
