Do you have a virtual machine running? (Parallels or VMware Fusion)

I had the same issue. Option + Eject seemingly did not work for my 2nd drive bay.

However, the issue started for me when I recently installed VMware Fusion and installed Windows XP onto it. I vaguely remember the setup asking me about drive setup and which one I wanted to use. Maybe I mis-configured something during the setup, but when I am running Windows XP via the VMware Fusion virtual machine, the drive in use for Windows cannot be ejected from the OS 10 side of things. Which makes sense when you think about it. Two SuperDrives, one available in each OS, when both are running.

I later figured out I can over-ride this option in VMware's settings, so that both drives are available on the Mac side.

Anyway, for me it was basically understanding how my drives are shared between Mac and Windows running in a virtual machine.
