I want to make an ssd as a primary drive for my mid 2010 iMac (11.3)! Will it work faster if i place it in an enclosure on the firewire port, or install it inside the iMac in the Sata3 port? (Like shown in the OWC video)

SATA III is gonna be much faster than FW 800, no contest. SATA III maxes out at 6 Gbps while FW 800 is of course 800 Mbps.

Ok, btw are we certain that my iMac inside has a Sata3 port or might it be a Sata2?

So SSD on SATA II internally for boot/OS/apps and FW800 external drives for large libraries and Time Machine backup (unless you are using something like a Time Capsule or NAS).
