Do I need to update the firmware on my Mac Pro 5,1 that currently has 138.0.0.0.0. To 140.0.0.0 to use a Samsung m.2 in PCIE as my boot drive?

Yes. MP5,1: BootROM thread | 140.0.0.0.0

Only if it's an NVME M.2. AHCI, Samsung SM951 AHCI and Samsung XP941, will boot without the 140 firmware. However the 140 firmware is recommended.
