    Today I replaced Apple's 4x 4GB RAM modules and installed 64GB purchased from OWC's (4x 16GB modules) into our new MP6,1 (6core, 1TB SSD, Dual D700s).

    It took 3 attempts.

1st attempt showed only 32 GB
2nd attempt showed only 48 GB
3rd attempt showed the full 64 GB.

In each case the issue was that the inner most 16GB module wasn't seated fully. It sure was tricky and not wishing to place too much pressure on the modules as I inserted them quite likely gave rise to the inner modules not being securely seated. The issue seemed to be the lower section of the inner modules needed a bit more pushing to fully seat.

I did run Geekbench 3 (64-bit) Version 3.1.5 (343) with the stock Apple 16GB RAM and scores were

    Single-Core: 3627
    Multi-Core: 20699

Running the Geekbench 3 (64-bit) Version 3.1.5 (343) with the new OWC's 64GB RAM the scores were

    Single-Core: 3679
    Multi-Core: 21060

Thus the RAM upgrade did improve on the scores some... :)

So here's a question, my System Information says I also have the 6.1 with the following specs:

    Model Name: Mac Pro
    Model Identifier: MacPro6,1
Processor Name: Quad-Core Intel Xeon E5
    Processor Speed: 3.7 GHz
    Number of Processors: 1
    Total Number of Cores: 4

I too want to upgrade the RAM at some point to 64 Gb and wonder if I would use the same spec RAM?

Just visit OWC's site and navigate to Memory. Then select the MP6,1 model to see the memory upgrade specs and prices.
