Good Day to all masters!

My jb ipad1 was mistakenly upgraded to 4.3.1 via itunes and could not access an apps which has data for our pregnancy workout of my wife.

Could I possibly restore from backup the data of the application which we need after jailbreak of 4.3.1. How is it done?

Please help me; data is very important for us and our doctor.

no. you cant go back unless you have your (insert correct name) key backed up with some "tiny umbrella" program.
 
even if i restore jb to 3.2.2? which is my previous jb OS

i do see my last back up file in
C:\Documents and Settings\Gian\Application Data\Apple Computer\MobileSync\Backup\cc48c5811395037dfb3247e147e6aee6d1ffb3e2
 
In theory if you dock your iPad, (plug it into your PC) and right click on the name in iTunes you should see an option to restore your iPad, this should in theory restore all data for app store apps, (that support this feature), at this point this is your best option, I think...:)

you should be able to update — even jailbreak — and keep all data. Specially since you made a backup on iTunes before upgrading. Or else, purchase the app "again" once you upgrade.

As @Dav1 says; "restore your iPad, this should in theory restore all data"
