How can I tell the difference between a MPEG-4 video file and a H.264 video file?

You can open it in Quicktime and then view the Film Info in the Window menu.

You can also find out simply via 'Get Info' (command + I) and checking the 'Codecs:' field under the 'More Info:' section.
